Akkermansia muciniphila Promotes Intestinal GLP-1 Secretion to Regulate Metabolic Homeostasis

2026-08-13

Akkermansia muciniphila, a key mucus-colonizing gut commensal bacterium, has emerged as a critical probiotic candidate for improving metabolic disorders. A growing body of peer-reviewed studies confirms thatA. muciniphila ameliorates glucose metabolism and insulin resistance mainly by stimulating the secretion of glucagon-like peptide-1 (GLP-1) from intestinal L-cells. This article summarizes core mechanisms and published experimental data to elaborate its metabolic regulatory function.

GLP-1 is an essential incretin hormone that enhances glucose-dependent insulin secretion, suppresses glucagon release, and inhibits excessive food intake, serving as a pivotal target for treating type 2 diabetes and obesity. Multiple authoritative studies have verified the positive correlation between A. muciniphila abundance and endogenous GLP-1 levels in the human and animal intestine.

A landmark study published in Nature Microbiology identified a specific functional protein P9 secreted by A. muciniphila, which directly binds to intestinal epithelial receptors to trigger GLP-1 transcription and release. In high-fat diet (HFD) mouse models, supplementation of viable A. muciniphila significantly upregulated intestinal GLP-1 expression, reduced fasting blood glucose, and effectively improved glucose homeostasis (Yoon et al., 2021). Cell experiments further proved that A. muciniphila extracts could induce a more than 20-fold increase in GLP-1 secretion from human intestinal L-cells in a dose-dependent manner (Kim et al., 2023).

In addition to specific proteins, the metabolic products of A. muciniphila also participate in GLP-1 regulation. The bacterium degrades intestinal mucus to produce short-chain fatty acids (SCFAs), especially acetate and propionate, which activate GPR41/43 signaling pathways on L-cell surfaces and further promote sustained GLP-1 secretion (Frontiers in Microbiology, 2025). Notably, clinical trials demonstrated that pasteurizedA. muciniphila still retains biological activity, increasing postprandial plasma GLP-1 levels by approximately 27% in overweight adults and improving peripheral insulin resistance (Plovier et al., 2021).

In conclusion, Akkermansia muciniphila promotes GLP-1 secretion through dual pathways of specific secretory proteins and microbial metabolites. It stably improves glycolipid metabolism in both animal models and human trials. With clear molecular mechanisms and reliable experimental data, A. muciniphila has great application potential in the development of functional foods and probiotic preparations for metabolic regulation.
